Colchicine Delayed the Experimental Liver Fibrosis in Rats but Prednisolon Stimulate Fibrosis

Abstract
We investigated the antifibrotic, effect of colchicine and prednisolon by the measurement of PNIIIP and PNIVP in serum. N-terminal procollagen type III peptide (PNIIIP ; collagen synthesis-Marker) and N-terminal procallagen type IV peptide(PNIVP : collagen degradation-marker) were sescribed the ongoing parameter of fibrosis processing in liver. The rats experimental animal that operated sham for normal and to induced liver fibrosis by the operation of bile duct ligation/scission(BDL/S), and observed 4 weeks after operation with treatment of colchicine(col : 0.15§¶/§¸/day) and prednisoln(pred : 1§·/§¸/day).
In the BDL/S operated group showed the significantly(p$lt;0.05) elevated value of PNIIIP(control : 2.66¡¾1.8ng/§¢, BDL/S : 9.3¡¾2.4ng/§¢), PNIVP(control : 3.0¡¾1.9ng/§¢, BDL/S : 7.8¡¾1.5ng/§¢) in serum and hydroxyproline(control : 1.2¡¾0.2§·/liver, BDL/D : 5.46¡¾1.0§·/liver) in liver tissue in compare to rats of sham-operated group, the similar data in BDL/S and pred, but BDL/S and col group(PNIIIP : control : 3.03¡¾0.9ng/§¢, BDL/S : 7.4¡¾3.03ng/§¢, PNIVP : control : 5.0¡¾1.68ng/§¢, BDL/S : 14.3¡¾1.3ng/§¢, hyp ; control : 1.5¡¾0.3ng/liver, BDL/S : 3.5¡¾2.0§·/liver). The content of hyp in liver tissue was correlated significantly with PNIIIP, PNIVP in serum in BDL/S(r=0.80, r=0.81), and BDL/S and pred treated rats(r=0.77. r=0.84). but not correlated in BDL/S and col treated group(r=0.45, r=0.64). In BDL/S and BDL/S-pred treated rats group observed massive proliferation of bile duct, formation of portal-portal septum and micro nodul by the histological finding in liver.
In conclusion that col inhibit weakly fibrogenesis and deposition of collagen in liver tissue, but stimulate fibrolysis. The pied do not delay the processing of experimental liver fibrosis. Col is effective antifibrotic agent for experimental liver fibrosis. The serological. measurement of PNIIIP and PNIVP is useful parameter for liver fibrosis and antifibrotic effect monitoring.
